Specific PCR Primer and Probe Design Based on Spanning Networks

While the amount of molecular data that can be routinely generated continues to increase, advances in the methods of analysis to handle this quantity of data have not kept pace. One area of research which could greatly benefit from new approaches is the design of specific probes and PCR primers. Here we generated spanning networks (1) for phytopathogenic Pseudomonas strains from seven species, based on GyrB and RpoD amino acid data. The spanning network approach is powerful except that this method does not provide accurate phylogenetic relationships when substitutions occur more than once at a site.
